白介素12(IL-12),也称为自然杀伤细胞刺激因子（NKSF）或毒性淋巴细胞成熟因子（CLMF），是一种多功能细胞因子，在EBV感染的RPMI-8866细胞培养液中被鉴定出的。IL-12是分子量为75kD的异二聚体组成的糖蛋白，其中，较小的亚单位(p35)与IL-6、G-CSF同源，而大亚单位(p40)被证明与可溶性IL-6受体有同源性。这提示IL-12可能是细胞因子与IL-6可溶性受体的复合物进化而来的。IL-12具有明显的种属特异性，人的IL-12与小鼠系统具有很少的交叉反应。巨噬细胞与B淋巴细胞表达的IL-12对T细胞与自然杀伤细胞有多种生理效应。包括诱导休止期与激活的T细胞、NK细胞表达IFN-γ与TNF，与其他IFN-γ协同诱导剂在转录与翻译水平上诱导IFN-γ表达，增强NK细胞与T细胞的杀伤能力，诱导被激活的T细胞与NK细胞分裂增殖。
产品原理
本实验采用双抗体夹心ELISA法。抗大鼠IL-12p70单抗包被于酶标板上，同时加入标本、标准品和生物素化的抗大鼠IL-12p70抗体，标本、标准品中的IL-12p70会与加入于孔中的生物素化的抗大鼠IL-12p70抗体及包被于酶标板上的单抗结合，形成免疫复合物，游离的成分被洗去；加入辣根过氧化物酶标记的亲合素，亲合素与生物素特异性结合，游离的成分被洗去。加入显色底物（显色剂），若反应孔中有IL-12p70，辣根过氧化物酶会使无色的显色剂变蓝，加终止液变黄。在450nm处测OD值，IL-12p70浓度与OD450值之间呈正相关，可通过绘制标准曲线求出标本中IL-12p70浓度。
